MIPEP also known as mitochondrial intermediate peptidase is a mitochondrial enzyme with a mass of approximately 78 kDa. This protein functions in the mitochondria where it processes precursor proteins by cleaving specific N-terminal residues after import into the organelle. MIPEP is expressed in a wide range of tissues but shows higher levels in the heart skeletal muscle and kidney. Its enzymatic activity is important for the maturation of mitochondrial proteins which are needed for proper mitochondrial function.
Biological function summary
MIPEP plays an important role in the processing and maturation of several mitochondrial matrix proteins. It acts in concert with other processing peptidases assisting in the final steps of protein maturation after the mitochondrial processing peptidase (MPP) cleaves the leader sequences from proteins. MIPEP works as part of a proteolytic system within the mitochondria that ensures proteins are in their functional forms. This processing ability is integral to maintaining the integrity and efficiency of the mitochondrial respiratory chain.
Pathways
MIPEP contributes significantly to mitochondrial protein import and processing pathways. The protein is involved in the maturation pathway of mitochondrial proteins which is critical for assembling electron transport chain complexes. MIPEP interacts with related peptidases such as MPP facilitating the process of protein maturation needed for efficient mitochondrial energy production. This interaction is essential for linking protein processing with energy metabolism.
Abnormalities in MIPEP function have been associated with combined oxidative phosphorylation deficiency. This disorder impairs energy production leading to a range of symptoms such as muscle weakness and neurological deficits. Additionally MIPEP dysfunction has links to mitochondrial diseases where it might affect proteins like MPP disrupting normal protein processing and exacerbating disease phenotypes. Understanding MIPEP's role may provide insights into the molecular basis of mitochondrial disorders and aid in developing therapeutic strategies.
