RABGGTB also known as Rab geranylgeranyltransferase subunit beta is an essential protein involved in the post-translational modification of Rab GTPases. This enzyme with a mass of approximately 37 kDa forms part of the larger Rab geranylgeranyltransferase complex. RABGGTB is expressed in various tissues throughout the body playing a critical role in cellular membrane trafficking processes. Rab GTPases need to be prenylated by RABGGTB for proper membrane attachment and functionality.
Biological function summary
RABGGTB is a part of the Rab geranylgeranyltransferase complex responsible for the dual prenylation of Rab proteins which include 'g' 'g' 't' 'b' '1' '6' and 'g' 'g' 't' 'b' '3' '7'. The prenylation facilitates the anchoring of Rab proteins to intracellular membranes which is essential for intracellular transport and signal transduction. Such modifications ensure that Rab GTPases act effectively in their roles maintaining vesicular traffic and cellular communication.
Pathways
RABGGTB contributes to essential pathways like vesicle-mediated transport and intracellular trafficking. It interacts closely with the small GTPase Rab family which significantly impacts these pathways. The correct functioning and localization of Rab GTPases activated by RABGGTB support key processes like endocytosis and exocytosis necessary for proper cell operation. Together with other proteins such as RABGGT1 RABGGTB anchors Rab GTPases aiding in the efficient execution of vesicular transport pathways.
RABGGTB has connections with disorders like Choroideremia and Hermansky-Pudlak syndrome. These conditions often link with defects in Rab protein prenylation illustrating the impact of RABGGTB's function on visual and pigmentary system health. The absence or dysfunction of RABGGTB disrupts normal Rab GTPase operations showing its association with the pathophysiological characteristics of these disorders. Notably it interacts with other proteins such as CHM influencing disease mechanisms by affecting Rab protein prenylation.
