Product details:
Abcam's PDGF-AA Rat ELISA (Enzyme-Linked Immunosorbent Assay) kit is an
in vitro
enzyme-linked immunosorbent assay designed for the quantitative measurement of rat PDGF-AA in serum, plasma, and cell culture supernatants.
This assay employs an antibody specific for rat PDGF-AA coated on a 96-well plate. Standards and samples are pipetted into the wells and PDGF-AA present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-rat PDGF-AA antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of PDGF-AA bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

PDGF-AA also known as Platelet-Derived Growth Factor AA is a protein with an approximate molecular mass of 28 kDa. It belongs to the PDGF family and functions as a significant growth factor. PDGF-AA is secreted as a homodimer and is mainly expressed in cells such as fibroblasts and smooth muscle cells. It interacts through PDGF receptors especially PDGFR-alpha which mediates its signalling activity. This interaction triggers various cellular responses essential for development and repair processes.
Biological function summary
PDGF-AA influences numerous cellular mechanisms including cell proliferation migration and angiogenesis. It acts as a powerful mitogen for mesenchymal cells being part of a dimeric complex. It also supports the maturation and differentiation of these cells. The presence of PDGF-AA is often important for the wound healing process and tissue regeneration playing an essential role in restructuring tissues after injury.
Pathways
PDGF-AA participates actively in the PI3K/Akt and MAPK signaling pathways. These pathways aid cellular functions related to growth survival and division. PDGF-AA activation can significantly impact these pathways by initiating their signalling cascades. The protein connects to other critical proteins like PI3K and Akt through these pathways contributing to broader cellular activities important for effective cellular responses.
PDGF-AA has strong ties to diseases such as atherosclerosis and certain cancers. These conditions often involve abnormal cell proliferation and migration processes in which PDGF-AA plays a critical role. Research shows that anomalies in PDGF-AA signalling might link to an increased risk of cancer progression. Moreover the protein has connections to PDGF receptors like PDGFR-alpha in the development and pathological progression of these diseases highlighting its significance in medical research and potential therapeutic targeting.
