Myosin light chain often referred to as MLC or the light chain myosin plays an important role in muscle contraction by binding to myosin heavy chains. This protein has a molecular mass of approximately 20 kDa and exists in smooth and striated muscle tissues as well as some non-muscle cells. MLC participates in regulating muscle contraction by interacting with myosin head groups during the ATPase activity which influences the actin-myosin interactions essential for muscle contraction.
Biological function summary
Myosin light chain interaction with myosin heavy chains regulates muscle contraction and relaxation. This small protein is a component of the larger myosin II complex which functions by modulating the motor activity on actin filaments. The phosphorylation state of the myosin light chain is critical in controlling the binding affinity of myosin to actin therefore directly affecting muscle contraction and cellular movement. In smooth muscles its phosphorylation by myosin light chain kinase increases the myosin ATPase activity contributing to muscle contractility.
Pathways
The action of myosin light chain fits into essential signaling processes regulating muscle function and cell motility. One significant pathway is the Rho/Rho-kinase pathway where phosphorylation of the myosin light chain modulates the contraction of smooth muscle and influences cell shape changes. Additionally the calcium-Calmodulin-Myosin Light Chain Kinase pathway also affects its phosphorylation status impacting smooth muscle contraction. Proteins such as phosphomyosin and myosin phosphatase interact within these pathways reinforcing the functional versatility of myosin light chain.
