GPR161 also known as G protein-coupled receptor 161 is a large transmembrane protein with a molecular weight of approximately 121 kDa. It belongs to the family of G protein-coupled receptors (GPCRs) and is mainly expressed in cilia of various cell types especially in the neural tube and limb buds during development. This receptor engages in the modulation of intracellular signaling pathways by sensing extracellular molecules.
Biological function summary
GPR161 functions as a negative regulator of the hedgehog (Hh) signaling pathway. It does so by promoting the degradation of GLI transcription factors in the absence of hedgehog ligands. GPR161 integrates within the ciliary membrane compartment playing an important role in maintaining proper cellular response to Hedgehog pathway signals. Its activity ensures balanced cellular growth and differentiation critical during embryonic development.

GPR161 acts significantly within the hedgehog signaling pathway and the primary cilia-related signaling processes. It correlates with other proteins like Patched1 and Smoothened which are essential in Hh signaling cascade.
