PILRB also known as paired immunoglobulin-like type 2 receptor beta is a transmembrane protein involved in immune signaling. It has a molecular weight of approximately 33 kDa. You can find its expression mainly on the surface of various immune cells including myeloid and natural killer (NK) cells. PILRB contains an immunoreceptor tyrosine-based activation motif (ITAM) within its cytoplasmic domain which plays a role in transmitting activation signals inside the cells.
Biological function summary
In immune response and signaling PILRB functions by interacting with other proteins on immune cells to modulate their activity. It works as part of a receptor complex with paired immunoglobulin-like type 2 receptor alpha (PILRA) which plays a complementary inhibitory role. This balance between PILRB and PILRA on cell surfaces helps regulate immune cell activation and maintain homeostasis within the immune system.
Pathways
PILRB has a role in pathways related to immune regulation and innate immunity. It participates in signaling pathways that involve its counterpart PILRA. An important pathway where PILRB is active includes the immune checkpoint pathway where it is related to NK cell activation and cytokine production. PILRB modulates the activity of other signaling molecules and receptors playing a part in pathways shared with proteins like SHP-1 and SHP-2.
Studies have shown PILRB's relevance in autoimmune diseases and cancer. Alterations in PILRB signaling can influence the progression of autoimmune conditions by affecting immune cell activation and self-tolerance. It is also involved in tumor immune evasion mechanisms. In these contexts PILRB relates to proteins like PD-1 and CTLA-4 which are also critical in immune checkpoint pathways and associated with both autoimmune diseases and cancer immunotherapy targets.
