CAPNS2 also known as Calpain Small Subunit 2 or CSS2 is a protein that is part of the calpain family a group of calcium-dependent non-lysosomal cysteine proteases. It has a molecular mass of approximately 29 kDa. CAPNS2 plays a role in intracellular signaling processes by mediating the cleavage of various substrates. It is expressed in numerous tissues with higher levels observed in skeletal muscle and brain reflecting its diverse physiological roles.
Biological function summary
CAPNS2 contributes to the regulation and modulation of cytoskeletal remodeling and cell motility. As a component of the calpain holoenzyme complex it partners with the large catalytic subunits CAPN1 and CAPN2 facilitating the calpain's proteolytic activity. This relationship highlights CAPNS2's role in enabling precise and controlled cellular processes such as cell cycle progression and signal transduction.
Pathways
CAPNS2 integrates into the calcium signaling pathway and is involved in the regulation of the cytoskeleton. In the calcium signaling pathway it interacts with CAPN2 participating in the activation of downstream effectors that convey calcium's pivotal roles in cellular functions. Additionally through its engagement in cytoskeletal reorganization CAPNS2 influences cell shape and migration playing a part alongside proteins like filamin and talin.
CAPNS2's malfunction or deregulation associates with neurodegenerative diseases and muscular dystrophies. In neurodegenerative conditions such as Alzheimer's CAPNS2 interacts with tau protein where dysregulation may contribute to neuronal cell death. Its role in muscular dystrophies links to the impaired function of related proteins like dystrophin underlining the importance of CAPNS2 in maintaining muscle integrity and function.
