Protein tyrosine phosphatase non-receptor type 22 (PTPN22) also known as Lyp is a phosphatase enzyme with a molecular mass of approximately 98 kDa. It removes phosphate groups from specific proteins participating in the regulation of signaling pathways that control cellular processes. PTPN22 is expressed in lymphoid tissues including T-cells B-cells and other immune cells. This distribution indicates its role in immune system function and signaling.
Biological function summary
PTPN22 plays a role in modulating immune responses impacting the activation of T-cells and B-cells. By altering signal transduction pathways it affects the threshold for immune cell activation. PTPN22 interacts with other signaling molecules but it is not known to be a part of any well-described protein complex. Its activity influences immune tolerance and autoimmunity making it an important player in maintaining immune system balance.
Pathways
PTPN22 is involved in the T-cell receptor signaling and negative regulation pathways. It dephosphorylates lymphocyte-specific protein tyrosine kinase (LCK) and ZAP-70 which are pivotal in T-cell activation. By regulating such proteins PTPN22 plays a direct role in modulating T-cell responses. The signaling balance achieved through its actions prevents overactive immune responses that could lead to autoimmune conditions.
PTPN22 has strong connections to autoimmune diseases such as rheumatoid arthritis and type 1 diabetes. Variants in the PTPN22 gene particularly the R620W allele are associated with an increased risk for these conditions. This variant affects the interaction of PTPN22 with C-SRC tyrosine kinase altering the normal immune signaling and contributing to the development of autoimmunity.
