TCR V gamma 9 also known as T cell receptor gamma 9 plays an important role in the immune system by recognizing antigens presented by non-classical MHC molecules. Measuring approximately 60 kDa TCR V gamma 9 forms part of the heterodimeric T cell receptor complex. It is particularly expressed on a subset of gamma delta T cells known as V gamma 9V delta 2 T cells which circulate in the blood and are involved in immune surveillance.
Biological function summary
TCR gamma 9 is involved in immune response to phosphoantigens derived from microbes and host metabolism. This receptor is part of a complex that includes the CD3 receptor protein which is required for signal transduction and T cell activation. The unique ability of V gamma 9V delta 2 T cells to recognize non-peptide antigens makes them different from the more common alpha beta T cells and positions them as important mediators in recognizing pathogen-induced stress.
Pathways
TCR V gamma 9 engages in immune cell signaling pathways such as the T-cell receptor signaling pathway and the MAPK signaling pathway. In these pathways it interacts with proteins like CD3 and ZAP-70 to facilitate T cell activation and proliferation. These interactions help initiate an immune response leading to the destruction of infected or cancerous cells.
TCR V gamma 9 has a role in infectious diseases like tuberculosis and certain cancers such as lymphoma. This receptor's interaction with CD3 plays a part in the host defense against Mycobacterium tuberculosis by recognizing unique phosphoantigens presented by this pathogen. Additionally alterations in gamma delta T cell function where TCR V gamma 9 is a core component correlate with cancer progression pointing to potential therapeutic interventions targeting this pathway in oncology.
