Wnt3 and Wnt3a both members of the Wnt family are secreted glycoproteins involved in several signaling pathways. Commonly known as Wnt3 and Wnt3a for simplicity they have an approximate mass of around 40 kDa. These proteins exhibit expression during development stages and in various adult tissues. They function mechanistically by binding to Frizzled family receptors activating downstream signaling cascades. Wnt3 is predominantly expressed in the central nervous system while Wnt3a shows wide expression across mesoderm and ectoderm tissues.
Biological function summary
Wnt3 and Wnt3a are key players in cell proliferation differentiation and migration. These proteins are part of the Wnt signaling complex which influences gene expression important for embryonic development and tissue homeostasis. The Wnt3 and Wnt3a promote stem cell maintenance and expansion by regulating transcriptional activators and repressors. Their role extends to maintaining cellular architecture through interaction with cytoskeletal elements.
Pathways
Wnt3 and Wnt3a are integral components of the canonical Wnt/β-catenin pathway and the Wnt/Ca2+ non-canonical pathway. Through these pathways they impact various biological processes essential to cellular growth and differentiation. Wnt3a often interacts with β-catenin a well-known mediator that influences gene expression by entering the nucleus. Both Wnts regulate pathways that include Dishevelled (Dvl) proteins highlighting their interconnectedness in cell signaling networks.
Alterations in Wnt3 and Wnt3a signaling contribute to cancer and neurodegenerative diseases. Abnormal Wnt/β-catenin signaling links to colorectal cancer where excessive β-catenin activity supports tumor growth. Moreover disrupted Wnt signaling can impact Alzheimer's disease pathogenesis through interactions with amyloid precursor protein and tau. These connections highlight how Wnt3 and Wnt3a influence disease states impacting therapeutic strategies targeting Wnt pathway components.
