ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative determination of autoantibodies to thyroid peroxidase in Human serum or plasma (citrate).
A 96-well plate has been precoated with recombinant thyroid peroxidase to bind cognate antibodies. Controls or test samples are added to the wells and incubated. Following washing, a horseradish peroxidase (HRP) labelled anti-Human IgG conjugate is added to the wells, which binds to the immobilized thyroid peroxidase-specific antibodies. TMB is then catalyzed by the HRP to produce a blue color product that changes to yellow after adding an acidic stop solution. The intensity of yellow coloration is directly proportional to the amount of thyroid peroxidase sample captured in plate.

Anti-Thyroid Peroxidase targets the enzyme Thyroid Peroxidase (TPO) which is essential in the synthesis of thyroid hormones. Also known as thyroperoxidase this protein has a mass of approximately 105 kDa. Thyroid Peroxidase is mainly expressed in the thyroid gland where it catalyzes the iodination of tyrosine residues in thyroglobulin a critical step in the production of the hormones thyroxine (T4) and triiodothyronine (T3).
Biological function summary
Thyroid Peroxidase facilitates the organification of iodine converting iodide ions into reactive iodine intermediates. It operates in conjunction with hydrogen peroxide provided by the enzyme Dual Oxidase (DUOX) forming a vital enzymatic complex within the apical membrane of thyroid follicular cells. This complex is necessary for the proper iodination of tyrosyl residues and subsequent coupling reactions that form T4 and T3 which regulate metabolism and normal growth.
Pathways
Thyroid Peroxidase plays an important role in the thyroid hormone synthesis pathway which is important for metabolic control and regulation of various physiological processes. It is closely linked with Thyroglobulin which serves as the substrate for iodination. Additionally it is part of the pathway that involves the Hypothalamus-Pituitary-Thyroid (HPT) axis maintaining homeostasis of thyroid hormones throughout the body via feedback mechanisms that involve proteins such as Thyrotropin-Releasing Hormone (TRH) and Thyroid-Stimulating Hormone (TSH).
Anti-Thyroid Peroxidase antibodies are markers of autoimmune thyroid diseases such as Hashimoto's thyroiditis and Graves' disease. These conditions cause the immune system to target Thyroid Peroxidase resulting in hypothyroidism or hyperthyroidism respectively. In these disorders the presence of Anti-Thyroglobulin can also be significant as it may indicate a concurrent autoimmune response impacting thyroid function.
