PSD95 (Postsynaptic Density Protein 95), encoded by gene DLG4, is a crucial scaffolding protein in neuro research, particularly for its role in synaptic stability and plasticity. It is involved in the organization of synaptic signaling complexes and the regulation of synaptic strength. Researchers use PSD95 to study synaptic development, neurotransmitter receptor localization and the effects of neurological disorders such as stroke and traumatic brain injury (TBI). Its expression helps in understanding synaptic function and the molecular mechanisms underlying synaptic plasticity and neuroprotection.

Protein PSD95 also known as postsynaptic density protein 95 or SAP90 is a significant component in synaptic function. This protein with a molecular mass of approximately 95 kDa is widely expressed in the central nervous system particularly in the postsynaptic terminals of neurons. PSD95 is a member of the membrane-associated guanylate kinase (MAGUK) family and contains three PDZ domains which facilitate its scaffolding ability to assemble large protein complexes at synaptic sites.
Biological function summary
Protein function contributes to the organization and regulation of synaptic signaling complexes. PSD95 is a core component of the postsynaptic density (PSD) an important structure for synaptic signaling and plasticity. The protein interacts directly with NMDA receptors and other synaptic proteins such as Shaker-type K+ channels which enhances synaptic strength and influences synaptic trafficking processes. These interactions are essential for the modulation of synaptic signals and the regulation of synaptic plasticity.
Pathways
Protein PSD95 integrates into several key neuronal signaling pathways. It plays a central role in the glutamatergic synapse as a scaffold protein facilitating receptor clustering and signaling transmission. Through interactions with NMDA receptors and AMPA receptors it modulates synaptic plasticity and long-term potentiation (LTP) critical processes in learning and memory. PSD95 is associated with proteins like stargazin and the guanylate kinase domain which influence synaptic dynamics through complex interactions.
Protein PSD95 plays a role in conditions like schizophrenia and Alzheimer's disease. Dysregulation of PSD95 is linked to impaired synaptic function and cognitive decline in these disorders. In schizophrenia abnormal PSD95 expression disrupts postsynaptic signaling leading to synaptic dysfunction. In Alzheimer's disease PSD95 interacts with amyloid-beta peptides contributing to synaptic deterioration. The protein establishes a connection with other proteins such as tau influencing neurodegeneration pathways.
