Alpha amylase also known as 14-alpha-D-glucan glucanohydrolase is an enzyme that catalyzes the hydrolysis of alpha bonds in large alpha-linked polysaccharides such as starch and glycogen breaking them down into glucose and maltose. The enzyme has a molecular mass that ranges from approximately 50 to 60 kDa depending on the isoform. Alpha amylase is expressed in the salivary glands and pancreas in humans where it initiates the digestive process. Additionally it is present in some microbiota aiding carbohydrate metabolism.
Biological function summary
Alpha amylase plays an important role in carbohydrate digestion breaking down dietary starch into simpler sugars for absorption. Although alpha amylase is not typically part of a larger protein complex its activity complements other enzymes like maltase and isomaltase in the digestive tract. By converting complex carbohydrates into absorbable glucose alpha amylase aids in maintaining energy homeostasis in the body influencing glucose metabolism and energy availability.
Pathways
Alpha amylase is critical in the digestive system's carbohydrate metabolism pathway. This pathway begins in the mouth and continues through the small intestine contributing to overall digestive efficiency. In the context of glucose metabolism alpha amylase works synergistically with enzymes such as glucokinase and glucose-6-phosphatase ensuring glucose levels are effectively managed for cellular energy production. This regulation is essential for maintaining normal blood sugar levels and supporting metabolic health.
Alpha amylase has been linked to conditions like diabetes and pancreatitis. In diabetes abnormal alpha amylase activity can contribute to dysregulated glucose levels complicating metabolic control. In cases of pancreatitis elevated levels of alpha amylase in the blood serve as a biomarker for the disease due to the enzyme's leakage from damaged pancreatic cells. Both conditions can involve interactions with proteins such as insulin in diabetes and trypsin in pancreatitis further influencing the disease mechanisms and progression.
