METTL21D also known as EF2-KMT or methyltransferase-like protein 21D is an enzyme with a molecular mass of approximately 33 kDa. It belongs to the protein lysine methyltransferase family and primarily acts on eukaryotic elongation factor 2 (eEF2) adding methyl groups to lysine residues affecting protein function. METTL21D expression is observed in various tissues including the brain and skeletal muscle indicating its widespread functional importance.
Biological function summary
METTL21D influences protein synthesis through its methyltransferase activity. It modifies eEF2 which is essential for the elongation phase during protein translation. By modifying eEF2 METTL21D impacts protein translation rates and cellular protein homeostasis. While not part of a larger complex METTL21D's interactions with other proteins highlight its regulatory function in translation.
Pathways
Protein translation and cellular metabolism feature prominently in METTL21D's activity. Its modification of eEF2 integrates METTL21D into the translation pathway influencing global protein synthesis. METTL21D also connects to pathways involving protein quality control where it potentially interacts with other methyltransferases that regulate cellular stress responses.
METTL21D links to conditions such as neurodegenerative diseases and certain types of cancer. Alterations in its expression or function can disrupt protein homeostasis contributing to disease progression. METTL21D through its interaction with eEF2 and possibly other methyltransferases may therefore affect pathways that lead to the development of these disorders providing insights for potential therapeutic interventions.
