RSPO4 also known as R-spondin 4 is a protein that plays a role in cell signaling and regulates growth and development processes. Its molecular weight is approximately 22 kDa. RSPO4 is active in various tissues with expression in the intestine bone and other epithelial tissues. It functions by enhancing Wnt signaling an important mechanism for cell proliferation and differentiation through binding to LGR receptors which then interact with ZNRF3 and RNF43 to prevent the degradation of Wnt co-receptors.

It does not work alone; instead it is part of a complex that includes LGR4 LGR5 and LGR6 receptors which facilitates the amplification of Wnt signals.
