RSPO4 also known as R-spondin 4 is a protein that plays a role in cell signaling and regulates growth and development processes. Its molecular weight is approximately 22 kDa. RSPO4 is active in various tissues with expression in the intestine bone and other epithelial tissues. It functions by enhancing Wnt signaling an important mechanism for cell proliferation and differentiation through binding to LGR receptors which then interact with ZNRF3 and RNF43 to prevent the degradation of Wnt co-receptors.
Biological function summary
The role of RSPO4 involves the modulation of cellular communication essential for tissue development and regeneration. It does not work alone; instead it is part of a complex that includes LGR4 LGR5 and LGR6 receptors which facilitates the amplification of Wnt signals. This activity supports processes such as stem cell maintenance and epithelial tissue homeostasis.

RSPO4 functions as an important enhancer of the Wnt/β-catenin signaling pathway and also interacts with non-canonical Wnt pathways. Disruption in RSPO4 expression relates to developmental issues and certain types of cancers. RSPO4 mutations are known to cause anonychia congenita a condition characterized by the absence of nails.
