The c-Jun N-terminal kinases 1 and 3 (JNK1 and JNK3) are members of the mitogen-activated protein kinase (MAPK) family also known as Stress-Activated Protein Kinases (SAPKs). JNK1 has a molecular mass of approximately 46-54 kDa while JNK3 has a mass of about 54 kDa. JNK1 is expressed in most tissues with high levels in heart brain and leukocytes whereas JNK3 is mainly expressed in the brain but also found in the testes and heart. These kinases play a role in signaling cascades that respond to stress stimuli such as cytokines ultraviolet radiation and reactive oxygen species.
Biological function summary
JNK1 and JNK3 regulate transcription factors and influence gene expression to modulate cellular processes. They are part of a complex network that includes other proteins like MAP kinase kinases (MKKs) which activate JNKs through dual phosphorylation. JNK1 and JNK3 control processes such as cell proliferation apoptosis and differentiation. Their activation can trigger the phosphorylation of c-Jun a component of the Activator Protein 1 (AP-1) transcription factor complex influencing cell fate decisions.
Pathways
JNK1 and JNK3 are involved in important signaling pathways such as the MAPK signaling pathway and the apoptosis signaling pathway. These pathways interact with other proteins like MKK4 and MKK7 which act upstream to activate JNKs. The JNK pathway helps the cell respond to stress and regulate apoptosis interacting with proteins such as Bcl-2 which is involved in many cellular responses including cell survival and death.
JNK1 and JNK3 have been associated with conditions such as neurodegenerative diseases and cancer. Abnormal activation of these kinases is linked to Alzheimer’s disease which involves protein misfolding and synaptic dysfunction and to various cancers due to their role in controlling cell death and proliferation. In these contexts they interact with disease-related proteins like tau in Alzheimer's disease and p53 in cancer affecting disease progression and therapeutic responses.
