UBIAD1 also known as UbiA prenyltransferase domain-containing protein 1 is a human gene encoding an enzyme with an important role in cellular processes. This enzyme has a mass of approximately 32 kDa and is expressed in various tissues including liver kidney and heart. UBIAD1 is involved in the synthesis of menaquinone-4 (MK-4) a form of vitamin K2 and it participates in cholesterol metabolism. Through its function this enzyme contributes to the production of coenzyme Q10 essential for mitochondrial energy production.
Biological function summary
UBIAD1 helps regulate lipid metabolism and cellular oxidative stress responses. The protein does not form a larger complex but functions independently to facilitate the biosynthesis of specific lipids. Its role in managing oxidative stress is particularly important as it can impact the production of reactive oxygen species within cells. By maintaining the balance of cellular lipids and redox states UBIAD1 supports overall cell health.
Pathways
UBIAD1 acts in both the mevalonate pathway and the vitamin K cycle. In the mevalonate pathway UBIAD1 contributes to the production of cholesterol and non-sterol isoprenoids. It works alongside related proteins such as HMG-CoA reductase a critical enzyme in cholesterol biosynthesis. UBIAD1 also plays a role in the vitamin K cycle which is important for activating clotting factors necessary for blood coagulation. This involvement highlights its participation in key metabolic and regulatory pathways.
Mutations in UBIAD1 can lead to Schnyder corneal dystrophy (SCD) a rare genetic disorder characterized by progressive opacification of the cornea. Research indicates that abnormal UBIAD1 function can disrupt cholesterol metabolism resulting in crystalline deposits in the cornea. Additionally UBIAD1 has connections to cardiovascular diseases due to its role in cholesterol regulation. Proteins such as apolipoprotein E (ApoE) may interact with UBIAD1 in the context of these disorders influencing lipid transport and metabolism.
