Cyclin-dependent kinase 5 (Cdk5) is a type of protein kinase known mainly for its regulatory role in the nervous system. Cdk5 is activated upon binding to its activators p35 or p39 proteins or in some cases to a truncated version known as p25. The Cdk5/p35 complex plays a role in cellular signaling pathways with p35 having a molecular mass of approximately 35 kDa. Cdk5 is expressed in post-mitotic neurons but also found in other cells under specific conditions. These proteins maintain structural and functional stability within cells where they are expressed contributing to multiple physiological processes.
Biological function summary
Cdk5 with its activator forms a complex that is essential for neural development and function. The Cdk5/p35 or Cdk5/p25 complexes participate in neuron migration and axon guidance. When p35 cleaves into p25 it can lead to prolonged activation of Cdk5 causing dysregulation of cellular processes. This dysregulation affects cytoskeletal dynamics leading to alterations in synaptic function and signal transmission in neurons. Therefore Cdk5 helps cells to manage normal and necessary development processes in the brain and neurons.

The Ras-related protein Rab-3A and protein phosphatase PP1 are among proteins interacting with Cdk5 through signaling cascades. These connections highlight Cdk5's involvement in regulating synaptic vesicle exocytosis and cytoskeletal integrity. Such pathways demonstrate the importance of Cdk5 in maintaining neuronal functionality and connectivity.
Cdk5 particularly in its Cdk5/p25 form is linked to Alzheimer's disease and amyotrophic lateral sclerosis (ALS). In these disorders the aberrant activation of Cdk5 leads to tau hyperphosphorylation and neurofibrillary tangle formation events associated with Alzheimer's-related neurodegeneration. In ALS Cdk5 dysregulation contributes to motor neuron damage. The alteration from p35 to p25 is a critical factor in these pathologies interacting with tau protein which is connected to the progression of these degenerative conditions.
