ADCY8 also known as AC8 is a member of the adenylate cyclase family of enzymes. This enzyme catalyzes the conversion of ATP to cyclic AMP (cAMP) an important cellular signaling molecule. The molecular weight of ADCY8 is approximately 125 kDa. ADCY8 is primarily expressed in the brain particularly in areas such as the hippocampus cortex and cerebellum. It is also found in other tissues albeit in lower levels. As a membrane-bound enzyme ADCY8 activity is regulated by calcium and calmodulin which distinguishes it from other adenylate cyclase isoforms.
Biological function summary
The production of cAMP by ADCY8 plays a critical role in signal transduction pathways that mediate many physiological processes. ADCY8 operates as part of a larger signaling complex involving G-protein coupled receptors (GPCRs) which allows for dynamic regulation and integration of signals. Through cAMP ADCY8 influences processes such as synaptic plasticity learning and memory formation. Its activity can modulate the intracellular levels of calcium further affecting cellular responses.
Pathways
ADCY8 is a significant component in the cAMP signaling pathway where it regulates many downstream effects including protein kinase A (PKA) activation and cAMP responsive element-binding protein (CREB) phosphorylation. In addition ADCY8 links to the calcium signaling pathway as its activity is modulated by intracellular calcium levels. Through these pathways ADCY8 interacts with various proteins including calmodulin and PKA which are essential for translating extracellular signals into specific cellular responses.
Aberrant ADCY8 function has been implicated in neurological conditions such as depression and schizophrenia. These associations highlight the enzyme’s role in neurotransmitter-related signaling and brain function. ADCY8’s interaction with CREB and other proteins involved in the cAMP signaling pathway highlights its significance in mood regulation and cognitive processes. Understanding these connections may offer insights into potential therapeutic targets for these disorders.
