Spindlin 1 also known as SPIN1 is a protein that functions as a histone code reader. It weighs approximately 29 kDa and is highly expressed in the testes ovaries and certain cancer cell lines. Spindlin 1 has three tandem Tudor domains which allow it to interact with methylated histones specifically histone H3. These interactions enable SPIN1 to regulate gene expression by binding to modified histones and influencing the transcriptional machinery.
Biological function summary
SPIN1 acts as a regulator of gene expression by interacting with epigenetic markers on histones. It is part of the transcriptional regulation complex which includes other proteins involved in chromatin remodeling and gene activation. Its role in binding to trimethylated lysine 4 on histone H3 (H3K4me3) highlights its impact on transcriptional regulation processes. This interaction is important for maintaining the pluripotency of stem cells and possibly influencing cell differentiation.
Pathways
SPIN1 functions mainly within the epigenetic regulation and gene expression pathways. It affects the histone modification pathway and plays an important role in regulating the transcriptional activation process. SPIN1's relationship with other proteins like the histone methyltransferase SETD3 and components of the chromatin-remodeling complex defines its role in gene regulation mechanisms. Together these proteins determine chromatin structure and gene activity impacting transcriptional outcomes.
SPIN1 has associations with cancer particularly ovarian and testicular cancers. This link arises from its overexpression which can promote tumorigenesis by altering gene expression profiles in cancerous cells. SPIN1 also interacts with oncogenic pathways involving proteins like MYC further implicating it in cancer development. Additionally its abnormal functioning can contribute to infertility issues due to its vital role in reproductive tissues with links to proteins involved in spermatogenesis and oogenesis.
