Proteinase Activated Receptor 3 (PAR-3) also known as protease-activated receptor-3 is a member of the PAR family. It is a G-protein coupled receptor involved in thrombin signaling. Weighing about 42 kDa PAR-3 is expressed in various human tissues including platelets endothelial cells and several types of epithelial cells. Its expression is significant in the cardiovascular and nervous systems where it modulates different physiological responses.
Biological function summary
PAR-3 plays an important role in cellular responses such as inflammation wound healing and coagulation. This receptor is important for platelet activation by thrombin which is paramount in blood coagulation. Although not a part of any larger structural complex it works closely alongside other protease-activated receptors like PAR-1 and PAR-4 creating synergistic effects in many cellular pathways. The interplay between PAR-3 and anticoagulation pathways emphasizes its regulatory role in hemostasis.
Pathways
PAR-3 interacts significantly with the coagulation pathway and is a part of the signaling cascade that controls thrombin-mediated platelet activation. It also connects to the inflammatory response pathway where it cooperates with PAR-1 to mediate cellular responses that contribute to inflammation. These pathways link PAR-3 to other proteins including thrombin and various intracellular signaling molecules emphasizing its integrative function in cellular communication and homeostasis.
PAR-3 has correlations with cardiovascular diseases and cancer. In cardiovascular diseases its role in thrombin signaling associates it with thrombosis and related complications. In cancer aberrant PAR-3 expression could facilitate tumor progression and metastasis through inflammatory and coagulation pathways. Proteins like PAR-1 thrombin and various cytokines further connect PAR-3 to these pathological conditions highlighting the receptor's potential as a therapeutic target in disease management.
