PAX1 also known as paired box protein Pax-1 is a transcription factor with a mass of approximately 52 kDa. This protein features a paired domain that mediates DNA binding and plays a significant role in transcriptional regulation. PAX1 is expressed in the notochord presomitic mesoderm and connective tissues during embryonic development. It is important during pattern formation and particularly important in vertebral column and skeletal development.
Biological function summary
The paired box protein PAX1 functions as an important player in embryogenesis primarily influencing the formation and differentiation of tissues. It is not known to be part of any large protein complex. PAX1 regulates the expression of various genes involved in the development of the axial skeleton particularly in the vertebral column. The accurate expression of PAX1 ensures proper skeletal structure formation acting as a determinant in vertebrate development.
Pathways
PAX1 contributes significantly to the Hedgehog and Wnt signaling pathways both of which are indispensable in regulating cell growth differentiation and patterning during embryogenesis. PAX1 collaborates with other proteins like Gli3 an important transcription factor in Hedgehog signaling. Within these pathways PAX1 ensures proper signaling cascades that streamline developmental processes and morphology.
Alteration or mutation in the PAX1 gene associates with vertebral malformations and disorders such as scoliosis and Klippel-Feil syndrome. Studies indicate that disruptions in PAX1 expression can lead to these congenital abnormalities by affecting the protein's regulatory function in skeletal development. The protein Barx1 which also plays a role in skeletal development can show interconnected interactions with PAX1 highlighting the complexity of genetic regulation in these disorders.
