GJA3 also known as Connexin 46 (Cx46) is a gap junction protein essential for cell communication. The protein has a molecular mass of about 46 kDa. It forms intercellular channels that connect adjacent cells allowing for the transfer of ions and small molecules. GJA3 is mostly expressed in the lens of the eye where it plays an important role in maintaining lens transparency and homeostasis.
Biological function summary
GJA3 participates in the formation of gap junctions that are important for maintaining cellular processes in the eye lens. As part of a connexin family complex GJA3 interacts with other connexin proteins like Cx50 to ensure proper cell-to-cell communication. This interaction is vital for the metabolic cooperation between lens fibers and helps in maintaining the avascular nature of the lens.
Pathways
GJA3 contributes to the regulation of lens fiber cell differentiation and growth pathways. It closely interacts with the and phosphoinositide 3-kinase (PI3K) and calcium signaling pathways. These pathways are critical for lens development and for responding to stress within the lens. In conjunction with Cx50 GJA3 modulates these signaling pathways to regulate lens opacity and health.
Mutations in GJA3 associate with congenital cataracts which lead to vision impairment or loss. These cataracts often result from disrupted intercellular communication within the lens affecting lens transparency. Additionally alterations in GJA3 impact the function of Cx50 another lens-specific connexin and can exacerbate the severity of cataract formation. Understanding the GJA3 and Cx50 interaction gives insights into the pathological mechanisms behind lens-related disorders.
