MARVELD2 also known as Tricellulin is a tight junction protein with a molecular mass of approximately 64 kDa. It localizes mainly at tricellular tight junctions where three cells meet in epithelial tissues. Tricellulin forms a part of the structure that helps maintain the integrity of epithelial barriers and it is expressed in several tissues including the cochlea kidney and intestine. This protein plays an important role in forming a seal between cells by regulating paracellular permeability limiting the passage of molecules between cells.
Biological function summary
Tricellulin ensures the performance of tight junctions by sealing the spaces between epithelial cells. It functions as part of a large protein complex contributing to cellular adhesion and barrier functions. By interacting with other tight junction proteins such as occludin and claudins Tricellulin influences epithelial barrier integrity. Its proper operation is vital for preventing the unregulated passage of solutes across epithelial layers especially in tissues prone to fluid movement and ionic exchange.
Pathways
Several mechanisms involve Tricellulin to ensure cell layer cohesion and selective permeability. It plays an important role in the epithelial cell signaling pathway and connects to proteins such as ZO-1 influencing cell polarity and communication. The protein acts in pathways that regulate intracellular signaling important for maintaining healthy cell growth and differentiation in epithelial layers.
Tricellulin has a connection to nonsyndromic hearing loss and familial hypomagnesemia with hypercalciuria and nephrocalcinosis (FHHNC). Mutations in the gene encoding this protein can impair its function leading to epithelial barrier dysfunction contributing to these conditions. For nonsyndromic hearing loss Tricellulin function relates to other proteins like claudins which also have key roles in the cochlear barrier. Understanding the relationship between Tricellulin and these disorders helps to elucidate the underlying mechanisms that contribute to such conditions and the potential for targeting therapeutics.
