MATK also known as Megakaryocyte-associated Tyrosine Kinase is a non-receptor tyrosine kinase with an approximate mass of 72 kDa. This protein is mainly expressed in hematopoietic cells especially in megakaryocytes where it plays a significant role in cellular signaling. MATK shares structural similarity with the Csk (C-terminal Src kinase) family suggesting its involvement in regulatory processes within these cells. Its expression is not limited to hematopoietic tissues as it is also found in various other cell types indicating its wider cellular function.
Biological function summary
MATK participates in the negative regulation of Src family kinases (SFKs) by phosphorylating their inhibitory tyrosine residues. This action modulates the activity of SFKs and influences processes such as cell proliferation differentiation and migration. MATK is often involved in complexes with other signaling molecules playing a regulatory function in cellular events. Its exact role can vary depending on specific cellular contexts and signals.
Pathways
It is an important player in both the hematopoietic and immune cell pathways. In the hematopoietic pathway MATK regulates SFKs ensuring controlled cell division and differentiation. In the immune response its regulation of kinases affects how immune cells react to stimuli. Proteins such as LCK and Fyn members of the SFK family are directly impacted by MATK’s activity within these pathways highlighting its importance in cellular function.
Dysregulation of MATK is associated with certain leukemias where its impaired function leads to unchecked cell proliferation. Similarly improper regulation in the immune context can contribute to autoimmune diseases. Its relationship with Src family kinases particularly LCK and Fyn cements MATK's role in oncogenic processes and autoimmune conditions providing potential targets for therapeutic intervention.
