TAF8 also known as TAF2G or TBN is a protein involved in transcription regulation. It has a molecular mass of approximately 38 kDa. TAF8 mainly expresses in the cell nucleus where it is an essential component of the transcription machinery. The protein plays a mechanical role in assembling and stabilizing the structure of the TFIID complex which is critical for initiating RNA polymerase II-dependent transcription.
Biological function summary
TAF8 associates with other TFIID components to influence gene expression. It is a part of the TFIID multi-protein complex that mediates the binding of the transcription factor IID (TFIID) to specific promoters. This complex interaction allows TAF8 to facilitate the transcription of various genes. It also interacts with transcription factors and other co-activators playing an essential role in regulating cellular transcription machinery.
Pathways
TAF8 functions within the transcription initiation pathway and directly impacts the transcription of protein-coding genes. It works alongside proteins like TBP and other TAFs within this pathway to form a backbone for the general transcription machinery. Its interactions help regulate the accessibility of DNA to RNA polymerase II and modulate the transcriptional activity of several important genes in the cell.
Defects in pathways involving TAF8 have links to certain developmental abnormalities and transcription-related disorders. For example mutations affecting TAF8 can contribute to diseases such as certain cancer types where precise control of transcription is disrupted. TAF8's connection with other TAF proteins within the TFIID complex accentuates its role in maintaining normal cellular function and preventing transcription-related pathologies.
