Neurotensin Receptor 1 also known as NTSR1 functions as a G-protein coupled receptor (GPCR) that binds to the neuropeptide neurotensin. This protein consists of seven transmembrane domains typical of GPCRs and has a molecular mass of roughly 46 kDa. NTSR1 commonly expresses in the central nervous system particularly in the brain regions such as hypothalamus and basal ganglia and also in peripheral tissues like the gastrointestinal tract.
Biological function summary
NTSR1 influences signaling cascades that regulate various physiological processes. It forms part of a receptor-ligand complex with neurotensin a peptide that modulates neurotransmission and hormone regulation. Neurotensin and NTSR1 interaction can affect neurotransmitter release modulating pathways related to pain locomotion and thermoregulation. Researchers have observed significant roles of the receptor in neural proliferation as well.
Pathways
NTSR1 acts significantly in the dopaminergic and phosphatidylinositol signaling pathways. In the dopaminergic system NTSR1 interacts with dopamine D2 receptors influencing dopamine-mediated behaviors and responses. In phosphatidylinositol signaling it regulates downstream effectors like protein kinase C impacting cellular responses. These pathways integrate NTSR1 into broader signaling networks that control critical neuronal functions and responses.
NTSR1 has connections to certain neurological and psychiatric disorders such as schizophrenia and Parkinson's disease. Abnormal NTSR1 signaling affects dopaminergic neurotransmission which highlights its role in these conditions. Altered interactions with proteins like the dopamine D2 receptor contribute to the pathological symptoms observed in these diseases. Research into NTSR1 and its pathways may reveal targets for therapeutic intervention.
