EEPD1 also known as Endo-Exonuclease/Phosphodiesterase-1 is involved in DNA repair processes. This protein shows endonuclease activity important for processing DNA by cleaving phosphodiester bonds at DNA damage sites. EEPD1 has a mass of approximately 78 kDa and is expressed in various tissues with notable expression in the testis. It contributes significantly to the maintenance of genomic stability.
Biological function summary
EEPD1 plays a significant role in the maintenance of DNA integrity by participating in the DNA damage response. It acts outside of large complexes directly recognizing and responding to DNA double-strand breaks to facilitate their repair. EEPD1 ensures efficient repair of such lesions which are critical for preventing mutations during DNA replication and under conditions of genotoxic stress.
Pathways
EEPD1's role intertwines with cellular pathways addressing DNA damage and repair specifically in homologous recombination and the alternative end-joining pathways. Within these pathways EEPD1 interacts with proteins such as Rad51 and BRCA1 which orchestrate the process of homologous recombination to repair DNA breaks accurately. Its placement in these pathways highlights its importance in preserving genomic sequences during cell division and stress.
EEPD1’s function connects to cancer and neurodegenerative disorders. Deficient EEPD1 activity may lead to an accumulation of unrepaired DNA contributing to genomic instability and cancer progression. In cancer its interaction with proteins like BRCA2 suggests its potential as a marker for tumor susceptibility and response to DNA-damage-inducing therapies. In neurodegenerative disorders malfunction in DNA repair can lead to neuronal cell death linking EEPD1’s role with proteins involved in neuroprotection against DNA damage.
