Inducible cAMP early repressor (ICER) also referred to as ICER is a transcriptional repressor that plays a role in regulating gene expression. ICER is one of several products generated from the cAMP-responsive element modulator (CREM) gene through alternative promoter usage. ICER consists of an estimated molecular mass of around 20-22 kDa depending on the variant. It notably expresses in endocrine tissues such as the pituitary gland and testis as well as in immune cells and the brain. The protein operates by binding to cAMP response elements (CRE) within DNA sequences inhibiting the transcription of certain genes.
Biological function summary
This protein influences gene expression regulation during cellular processes such as differentiation and proliferation. ICER lacks independent transcriptional activity and functions through dimerization with other transcription factors. Although not a part of complex machinery like RNA polymerase ICER affects transcriptional repression via interactions with partners involved in the CREB family. It serves in maintaining cellular homeostasis especially during feedback regulation of genes activated by cAMP signaling.
Pathways
ICER impacts intracellular signaling by being involved in the cAMP signaling pathway and has roles in regulating cellular responses to hormonal stimuli. This pathway affects various physiological systems including the cardiovascular and nervous systems. ICER often interacts with CREB (cAMP-response element-binding protein) within these pathways. CREB and related proteins modulate gene transcription in response to hormonal changes linking ICER's function to broader signaling networks.
ICER relates to cardiac hypertrophy and depression where dysregulated cAMP signaling contributes to pathogenesis. In cardiac hypertrophy ICER disrupts normal gene expression patterns within the heart possibly linked with CREB signaling disruptions. In depression altered ICER expression in brain tissue affects responses to neurotransmitter signaling changes also linking back to CREB and other transcriptional regulators. Understanding ICER's roles in these conditions helps grasp how it influences disease states through transcriptional control.
