MT-ND1 also known as mitochondrial ND1 or NADH dehydrogenase 1 is a subunit of the enzyme NADH: ubiquinone oxidoreductase commonly called Complex I. This protein has a mass of approximately 35 kDa. It locates in the inner mitochondrial membrane where it serves a role in electron transport. MT-ND1 helps catalyze the transfer of electrons from NADH to ubiquinone an important step in the mitochondrial respiratory chain.
Biological function summary
The function of MT-ND1 is vital for ATP production. It forms part of the Complex I assembly within the mitochondrial membrane. Complex I is the first enzyme in the electron transport chain and initiates the transfer of electrons which ultimately produces the proton gradient needed by ATP synthase to generate adenosine triphosphate (ATP). This energy conversion process is essential for cellular energy homeostasis.
Pathways
MT-ND1 is integral to the oxidative phosphorylation pathway which is an important process for energy production in cells. Through the electron transport chain MT-ND1 is linked to other mitochondrial proteins such as MT-ND2 and MT-ND3 as they form subunits of the same complex. These subunits collectively contribute to maintaining the efficiency and regulation of cellular respiration.
Defects in MT-ND1 are associated with mitochondrial diseases such as Leber's hereditary optic neuropathy (LHON) and mitochondrial encephalomyopathy. MT-ND1 mutations may disrupt the electron transport chain leading to decreased ATP production and increased reactive oxygen species. These conditions are also related to other components of Complex I including MT-ND4 and MT-ND6 further affecting the functioning of the mitochondrial respiratory machinery.
