GNA14 also known as Guanine nucleotide-binding protein subunit alpha-14 is a component of the G protein family. This protein has a mass of approximately 41 kDa. It primarily expresses in the liver adrenal glands placenta and spleen. As a G protein it plays a role in signal transduction as part of the G alpha subunit which is an important player in relaying signals from cell surface receptors to downstream effectors within the cell.
Biological function summary
GNA14 mediates signal transduction by coupling with cell surface receptors to activate downstream signaling pathways. It functions as part of the larger heterotrimeric G protein complex interacting with G protein-coupled receptors (GPCRs). These GPCRs stimulate GNA14 which then activates intracellular signaling paths involved in various cellular responses including growth differentiation and metabolism playing an important role in maintaining cellular homeostasis.
Pathways
GNA14 participates mainly in the phospholipase C (PLC) signaling pathway where it interacts with PLC to regulate calcium ion release within the cells. This ion release affects various cellular processes such as muscle contraction and neurotransmitter release. GNA14 also shares pathway involvement with proteins like GNA13 another member of the G alpha subunit family which can have overlapping or distinct physiological roles.
GNA14 mutations have associations with conditions like uveal melanoma and Kaposiform hemangioendothelioma. In uveal melanoma GNA14 mutations affect normal signaling pathways leading to uncontrolled cell proliferation. In these diseases GNA14 does not work alone but interacts with other proteins involved in cancer pathway dysregulations such as GNAQ and BRAF implicating a complex network of molecular interactions in the pathology of such disorders.
