ACMSD also known as 2-amino-3-carboxymuconate-6-semialdehyde decarboxylase is an enzyme with a mass of around 39 kDa. It plays an important role in the metabolism of tryptophan converting 2-amino-3-carboxymuconate-6-semialdehyde into alpha-amino-beta-carboxymuconate-epsilon-semialdehyde. ACMSD is expressed mostly in the liver and kidney tissues. It functions at a critical step in the kynurenine pathway which processes tryptophan into several metabolites.
Biological function summary
ACMSD regulates the flux of metabolites through the kynurenine pathway influencing the production of NAD+ and quinolinic acid. ACMSD converts intermediates in the pathway which prevents the excessive build-up of quinolinic acid a neurotoxin. ACMSD is not typically part of a larger enzyme complex but interacts functionally within the pathway components. Its expression and activity impact the levels of various metabolites affecting cellular functions and energy metabolism.
Pathways
The kynurenine pathway is where ACMSD fits significantly. In this pathway ACMSD influences the synthesis of NAD+ which is essential for energy production and cellular signaling. ACMSD links closely with other enzymes like kynurenine 3-monooxygenase and kynureninase. By controlling the balance between different metabolites ACMSD helps maintain homeostasis and prevents potential toxic effects from accumulated intermediates like quinolinic acid.
ACMSD attracts attention for its association with neurological conditions and inflammatory diseases. Disruptions in its functioning can lead to abnormal levels of quinolinic acid contributing to neurodegenerative diseases like Alzheimer's and multiple sclerosis. Additionally abnormal regulation of ACMSD can affect tryptophan metabolism impacting disorders like chronic kidney disease. Connections with proteins like indoleamine 23-dioxygenase provide a broader understanding of its role in disease contexts.
