PEAMT or phosphoethanolamine N-methyltransferase is an enzyme involved in phospholipid biosynthesis. This protein sometimes referred to as PEMT facilitates the conversion of phosphoethanolamine to phosphocholine donating methyl groups during the reaction. PEAMT has a molecular mass around 23 kDa. Expression of PEAMT primarily occurs in tissues with high phospholipid turnover such as the liver.
Biological function summary
Phosphoethanolamine N-methyltransferase plays a significant role in the biosynthesis of phosphatidylcholine a critical phospholipid component of cell membranes. PEAMT operates as a part of methylation processes converting phosphoethanolamine into phosphocholine through a multi-step enzymatic pathway. It does not belong to a larger protein complex but acts independently to ensure adequate phosphocholine supply which is vital for maintaining membrane integrity and function.
Pathways
PEAMT is an important participant in the CDP-choline pathway also known as the Kennedy pathway. This biological route is essential for phosphatidylcholine synthesis joining the efforts of enzymes such as choline kinase and CPT1. PEAMT's enzymatic activity ensures a steady supply of phosphocholine which ultimately integrates into phosphatidylcholine to maintain membrane structure. Its role closely connects to methylation pathways tying it with proteins involved in one-carbon metabolism.
PEAMT's activity shows a connection to liver diseases and metabolic disorders. Nonalcoholic fatty liver disease (NAFLD) often shows altered levels of phosphatidylcholine where PEAMT's altered function plays a part in its pathogenesis. Additionally metabolic syndrome shows correlation with altered phosphatidylcholine synthesis pathways where PEAMT can influence lipid profiles. Links between PEAMT and S-adenosylmethionine another key methyl donor highlight the enzyme’s impact on disorders involving methylation imbalance.
