CRB1 also known as Crumbs Homolog 1 is a protein with a mass of approximately 160 kDa. Mechanically CRB1 plays a critical role in the structural maintenance of retinal photoreceptor cells. It localizes predominantly to the apical membrane of these cells in the retina. CRB1 helps organize intercellular junctions and maintain cell polarity providing essential support to the retina's architecture and function.
Biological function summary
CRB1 maintains cell polarity and adhesion in photoreceptors which are critical for normal vision function. It is part of the CRB1 complex which includes other proteins such as CRB2 and CRB3. This complex operates within the retina and the brain maintaining the structural integrity necessary for cellular communication and overall tissue organization.
Pathways
CRB1 plays a role in the Hippo signaling pathway and the cellular polarity regulation pathway. In these pathways it interacts with proteins such as MPP5 and AMOT which are integral for regulating cell growth and maintaining tissue homeostasis. These interactions ensure proper functioning and survival of retinal cells preventing their degradation over time.
CRB1 mutations associate with retinal dystrophies including retinitis pigmentosa and Leber congenital amaurosis. These conditions lead to progressive vision loss due to the degeneration of photoreceptor cells. Mutations can lead to dysfunctional interaction with proteins like PALS1 and CRB3 disrupting the cell polarity and junction maintenance which ultimately affects the structural support in the retina.
