E3 ubiquitin-protein ligase RNF213 also known as mysterin functions as an E3 ubiquitin ligase in the ubiquitin-proteasome system. This large protein has a molecular mass of around 591 kDa. RNF213 mediates the ubiquitination of substrate proteins which marks them for degradation. This enzyme expresses mainly in endothelial and smooth muscle cells with some presence in other tissues. RNF213's role in protein degradation is essential for cellular regulation and protein turnover.

RNF213 plays a significant function in vascular homeostasis and cellular stress response. It does not typically form part of a larger protein complex but interacts directly with its substrates during ubiquitination. Its activity is important for managing cellular processes linked to cell survival under various stress conditions. Other proteins such as p53 which interact with RNF213 play roles in cellular response to stress indicating possible links to other types of vascular disorders.
