CENPM also known as CENTRIN-M or LRPR1 is a protein that plays an important role in cell division. It has a molecular mass of approximately 19 kDa and is central to the assembly and stability of the kinetochore a structure that mediates chromosome segregation during mitosis and meiosis. CENPM is expressed in dividing cells particularly in tissues with high proliferative activity such as bone marrow and the gastrointestinal tract. This protein's function is indispensable for proper chromosome alignment and segregation ensuring correct genetic information transfer during cell division.
Biological function summary
CENPM contributes to chromosome alignment by being an integral part of the CENP-A nucleosome-associated complex. This complex comprises several other centromere proteins essential for kinetochore structure and function. CENPM's interaction facilitates the recruitment and stabilization of other critical proteins at the centromere such as CENP-H CENP-I and CENP-K. By influencing centromere integrity CENPM helps maintain genomic stability and prevents chromosome missegregation which could lead to aneuploidy.
Pathways
CENPM plays a significant role in the mitotic cell cycle pathway particularly in the spindle checkpoint. This checkpoint ensures that all chromosomes are correctly attached to the spindle apparatus before anaphase proceeds. CENPM collaborates closely with proteins like CENP-C and Bub1 to delay cell cycle progression until errors are corrected preventing premature anaphase onset. CENPM also associates with the centromere assembly pathway contributing to the proper localization of kinetochore proteins necessary for accurate chromosome segregation.
Issues with CENPM expression or function can contribute to cancer and aneuploidy disorders. Abnormalities in CENPM levels or mutations may lead to improper chromosome segregation resulting in cells with abnormal chromosome numbers a hallmark of many cancers. CENPM dysfunction is often linked with defective kinetochore assembly and can show connections to other proteins like CENP-A and CENP-B in the context of cancer development. Understanding CENPM's role in these diseases is important for developing potential therapeutic strategies targeting cell division anomalies.
