EPPK1 also known as epiplakin is a large protein with a molecular mass of approximately 725 kDa. It is part of the plakin family and functions as a cytolinker protein. EPPK1 is fundamentally present in epithelial tissues and its expression is noted in various organs including the skin stomach and esophagus. This protein aids in the stabilization of cytoskeletal structures by crosslinking intermediate filaments within the cells.
Biological function summary
EPPK1 contributes to the structural integrity of epithelial cells by promoting filamentous network organization. It acts as a component in complexes that assemble the cytoskeleton's intermediate filament network significantly impacting cellular resilience and shape. EPPK1's interactions are important for maintaining cellular architecture and play an important role in cell adhesion processes.
Pathways
EPPK1 plays a significant role in both cytoskeletal organization and the cellular response to stress. It is part of pathways that involve keratin proteins which are necessary for maintaining epithelial scaffold resilience. Its interaction with proteins like desmoplakin further integrates EPPK1 into desmosome complex pathways that are essential for cohesion among epithelial cells.
Mutations or altered expression of EPPK1 have been linked to skin disorders such as epidermolysis bullosa and other keratin-associated conditions. This protein's anomalies might influence the desmosome machinery with possible interactions affecting other proteins like plakoglobin which is also associated with cellular adhesion integrity. Understanding EPPK1’s involvement in these conditions could pave the way for better therapeutic strategies.
