Talin 2 also known as TLN2 is a large cytoskeletal protein approximately 270 kDa in mass. It acts mechanically as an important player in linking integrins to the actin cytoskeleton supporting the structural integrity of cells. Talin 2 is expressed in various tissues including muscle and neuronal systems with expression patterns similar to Talin 1 but has distinct roles in cellular adhesion and signal transduction.
Biological function summary
Talin 2 plays a significant role in cell adhesion and migration processes. It is part of a complex that includes integrins and other cytoskeletal proteins facilitating the transmission of mechanical signals across the cell membrane. This protein aids in focal adhesion assembly ensuring proper cellular responses to external stimuli. Talin 2 contributes to stabilizing intracellular interactions which are essential for maintaining tissue structure and cell motility.
Pathways
Talin 2 is deeply embedded in integrin-mediated signaling and the cytoskeletal organization pathway. It modulates interactions between integrins and F-actin orchestrating actin dynamics and contributing to cellular adherence junctions. Talin 2 functions closely with proteins like vinculin which supports actin filament linkage and reinforces mechanical stability under tension.
Talin 2 has been linked to muscular dystrophies and certain neuropathies. Mutations or dysregulation in Talin 2 can impair muscle function as its interaction with dystrophin is critical for muscle integrity. Additionally disruptions in Talin 2 mediated signaling could contribute to neurodegenerative disorders where its interactions with integrins can affect neuronal connectivity and survival. Understanding Talin 2's role in these conditions may provide insights for therapeutic interventions.
