Eph receptor B3 also known as Ephrin type-B receptor 3 or EphB3 is a member of the Eph receptor family. These receptors are important elements in receptor tyrosine kinases notable for their involvement in numerous cellular processes. EphB3 has a molecular mass of approximately 110 kDa. EphB3 is predominantly expressed in the brain but also found in lungs intestines and spinal cord. Its expression pattern hints at its diverse roles in various organ systems.
Biological function summary
Eph receptor B3 plays important roles in cellular communication influencing cell adhesion migration and differentiation. It often acts by forming receptor complexes with its ligands the ephrins. These interactions initiate bidirectional signaling which regulates the positioning of cells in developing tissues. EphB3 is important in guiding axon pathfinding in neurons and vascular patterning during development.

Eph receptor B3 signaling influences the Rho family GTPase pathway. This pathway is involved in cytoskeletal dynamics and cellular morphology changes. EphB3 also partakes in the MAPK/ERK pathway affecting cell proliferation and survival. Through the MAPK pathway EphB3 is linked with related proteins like EphA2 and EphB2 which share overlapping functions in cellular development and differentiation.
Eph receptor B3 has connections to cancer progression and neurological disorders. Changes in EphB3 expression levels have associations with colorectal cancer affecting cell invasion and metastasis. Additionally altered EphB3 signaling is reported in Alzheimer's disease where it seems to interact with the Amyloid precursor protein (APP) contributing to neurodegenerative processes. These associations highlight EphB3's importance in understanding and potentially targeting these conditions.
