Guanylyl cyclase-activating protein 2 (GCAP2) also known as GUCA1B is a calcium-binding protein with a molecular weight of approximately 24 kDa. GCAP2 plays an important role in the visual system's phototransduction process. It is primarily expressed in the retina especially in photoreceptor cells. GCAP2 is part of a family of neuronal calcium sensor proteins that adjust their activities based on calcium concentration changes. These adjustments help regulate the sensitivity of photoreceptors to light.
Biological function summary
This protein modulates the activity of retinal guanylyl cyclases. In photoreceptors it forms complexes with these enzymes to regulate the synthesis of cyclic GMP in response to light. The availability of cyclic GMP is essential for the recovery phase of phototransduction which allows visual cells to respond to light changes efficiently. GCAP2 switching between active and inactive states adjusts the production of cyclic GMP depending on calcium levels inside the cells.
Pathways
GCAP2 features in the phototransduction cascade an essential pathway for converting light into visual signals. This cascade involves several proteins including rhodopsin transducin and phosphodiesterase. GCAP2’s role focuses on regulating guanylyl cyclases allowing the restoration of cyclic GMP levels which is important for returning ion channels to their open state after photon absorption. It is tightly connected to GCAP1 another guanylyl cyclase-activating protein with similar functions but they operate under slightly different calcium concentrations.
GCAP2 mutations have been linked to certain forms of cone and cone-rod dystrophies retinal degenerative disorders that affect vision. Changes in GCAP2 function influence cyclic GMP levels disrupting normal photoreceptor activity and survival. Mutations can interact with the rhodopsin promoter influencing disease progression and severity. Understanding GCAP2’s role provides potential targets for treating these retinal disorders and improving photoreceptor health.
