MED7 also known as Mediator Complex Subunit 7 is a protein component of core Mediator complex essential for transcription regulation by RNA polymerase II. MED7 weighs approximately 25 kDa and exists in nucleus. It is part of a large multi-protein assembly. This subunit participates specifically by aiding in the mediator's interactions with gene-specific regulatory proteins and general transcription factors essential for transcription initiation.
Biological function summary
These components coordinate as part of Mediator complex which regulates transcription by acting as bridge between DNA-bound transcriptional regulators and RNA polymerase II. MED7 contributes to the structural integrity of Mediator complex affecting transcriptional activation processes necessary for gene expression. The protein's roles link it directly to cell growth differentiation and response to various stimuli indicating its importance in diverse cellular processes.
Pathways
MED7 integrates itself into transcription regulation pathways especially interacting with other subunits like MED1 and MED14. The Mediator complex functions in the pathway responsible for transferring information from transcription factors to RNA polymerase II. This involves interacting with multiple pathways including Notch signaling and Wnt signaling serving as coordinator between transcriptional regulators and transcription machinery.
MED7 has associations with cancer and neurodevelopmental disorders. Aberrations in Mediator complex components including MED7 can impact transcriptional dysregulation linked to oncogenesis. Mutations in this subunit along with changes in associated proteins such as MED12 are observed in various cancer types. Additionally disturbances in Mediator complex function potentially involving MED7 contribute to neurodevelopmental conditions due to misregulation of gene expression essential for neural development.
