Mitofusin 1 (MFN1) also known as mfn1 is a GTPase protein involved in mitochondrial fusion with a molecular weight of approximately 84 kDa. This protein is expressed in various tissues most notably in skeletal muscle cardiac muscle and the nervous system. Mitofusin 1 plays a significant role in maintaining mitochondrial structure and function by facilitating the fusion of outer mitochondrial membranes which is essential for normal mitochondrial dynamics.
Biological function summary
Mitofusin 1 is involved in processes beyond just mitochondrial fusion. It participates in mitochondrial quality control and bioenergetics helping to optimize the distribution of mitochondrial DNA and components across the mitochondrial network. Mitofusin 1 is also part of a complex that involves Mitofusin 2 (MFN2). Together these proteins regulate interactions between mitochondria and other organelles contributing to cellular homeostasis and adaptation to metabolic demands.
Pathways
Mitofusin 1 plays a role in the mitochondrial fusion and fission pathway which is critical for mitochondrial and cellular health. It interacts closely with other proteins like OPA1 (optic atrophy 1) an important regulator in the fusion process. Moreover Mitofusin 1 is integrated into the apoptosis pathway where it influences cell survival and death. Its interaction with Bak and Bax proteins which are involved in apoptotic signaling highlights the role of Mitofusin 1 in balancing cell fate decisions.
Mutations or dysregulation of Mitofusin 1 have associations with neurodegenerative diseases and cardiomyopathies. For instance defects in Mitofusin 1 are linked to Charcot-Marie-Tooth disease type 2A a disorder affecting peripheral nerves. Additionally disturbances in Mitofusin 1 function have implications for heart health particularly in relation to heart failure where proper mitochondrial dynamics are disrupted. Through these conditions Mitofusin 1 also interacts with associated proteins like Mitofusin 2 reflecting shared roles in the pathology of these disorders.
