EIF4E2 also known as eIF4E homologous protein (4EHP) is a mechanistic player in translation initiation. As a part of the eukaryotic translation initiation factor family it has a mass of approximately 25 kDa. EIF4E2 predominantly locates in the cytoplasm and its expression levels vary but often arise in specific tissues such as the brain and placenta. It interacts with other components of translation machinery acting to regulate protein synthesis by modulating mRNA cap binding activity.
Biological function summary
EIF4E2 plays a role in mRNA translation regulation. It forms a complex with other initiation factors such as eIF4G but lacks some binding capabilities of its closely related factor eIF4E. EIF4E2 impacts translation repression contributing by silencing specific mRNAs in cell cycles or stress responses. The function of EIF4E2 is necessary for maintaining cellular homeostasis particularly in adapting to environmental or cellular stresses.
Pathways
EIF4E2 participates significantly in the mTOR signaling and hypoxia response pathways. EIF4E2's involvement in mTOR signaling overlaps with its relationship to target proteins like 4E-BP1 which regulate cell growth and proliferation. In the hypoxia response pathway EIF4E2's translation control role is critical for cellular adaptation to low oxygen conditions by modulating hypoxia-inducible factors (HIFs).
Research links EIF4E2 to cancer and neurological disorders. Its role in abnormal cellular growth ties it to various cancers where changes in translation rates contribute to oncogenesis. EIF4E2's regulatory function in mRNA translation also connects it to neurodegenerative conditions which often involve dysfunctions in protein synthesis. EIF4E2 interacts with related proteins such as eIF4E and 4E-BP1 in these pathologies pointing to translation control as a potential therapeutic target.
