Alpha-synuclein often referred to as SNCA or NACP is a protein that plays important mechanical roles in the nervous system. It possesses a mass of approximately 14 kDa and is known for its expression predominately in the brain especially within the presynaptic terminals of neurons. Its primary function involves binding to phospholipids and regulating synaptic vesicle trafficking affecting the release of neurotransmitters. Alpha-synuclein can also adopt multiple conformations including monomeric and aggregated forms which can affect its functional capabilities.
Biological function summary
Alpha-synuclein impacts several cellular processes including neuronal function and plasticity. It can form aggregates known as Lewy bodies which are distinctive in certain neurodegenerative diseases. Alpha-synuclein is associated with the regulation of dopamine release and uptake affecting synaptic activity and neural communication. Within cells it does not necessarily function as part of a larger protein complex but interacts dynamically with cellular membranes and other synaptic proteins impacting cellular homeostasis and stress response.
Pathways
This protein integrates into key neuronal processes such as the synaptic vesicle cycle and the autophagy-lysosome pathway. It affects interactions with other proteins like PARK7 (DJ-1) and LRRK2 which influence neurodegenerative processes.
