UVRAG also known as UV radiation resistance-associated gene is a protein with a molecular weight of approximately 70 kDa. This protein exists within various tissues but shows higher expression in the brain heart and skeletal muscle. The mechanistic role of UVRAG involves its interaction with cellular membranes mediating autophagy and endocytic trafficking processes which are essential for cellular homeostasis and defense against stress.
Biological function summary
UVRAG regulates important cellular processes contributing to autophagy by promoting the maturation of autophagosomes into lysosomes. It is an integral part of the PI3K complex working alongside other proteins such as Beclin 1. Through these interactions UVRAG aids cell survival under nutrient-deficient conditions by facilitating the degradation of damaged organelles and protein recycling.
Pathways
UVRAG's role extends to its involvement in the mTOR signaling pathway and the endocytic pathway. Within the mTOR pathway UVRAG interacts with proteins like Rubicon and Beclin 1 acting as a modulator of cellular growth and response to energy status. In the endocytic pathway UVRAG collaborates with the VPS34 complex influencing the sorting and degradation of cellular cargo. These pathways highlight UVRAG's importance in cellular metabolism and regulation.
UVRAG shows significant connections particularly in cancer and neurodegenerative diseases. Mutations or altered expressions of UVRAG can contribute to tumorigenesis by disrupting autophagic balance or fail in cell death processes. In cancer UVRAG interacts with proteins such as p53 influencing tumor suppressor activities. In neurodegenerative disorders its regulatory role in autophagy becomes impaired contributing to protein aggregation and cell death highlighting UVRAG as a potential therapeutic target.
