GNB4 also known as G-protein subunit beta-4 is a component of the heterotrimeric G protein complex which plays a significant role in intracellular signaling. This protein has an approximate molecular weight of 39 kDa. GNB4 is primarily expressed in the brain and also detected in various tissues contributing to its diverse functional roles. As a β-subunit GNB4 forms part of a larger complex that mediates various signal transduction processes by interacting with different receptors on the cell surface.
Biological function summary
The functionality of GNB4 involves interactions within the heterotrimeric G protein complex a critical component of signal transduction pathways. This protein acts as a modulator or adapter in transmembrane signaling systems. GNB4 pairs with gamma subunits to help regulate downstream effector proteins expanding the impact of receptor-binding events initiated by external stimuli. GNB4 influences processes like cell communication and neurotransmission making it an important factor in maintaining cellular functions in neural networks.
Pathways
GNB4 plays essential roles in G-protein coupled receptor (GPCR) signaling and other signaling pathways. In GPCR signaling GNB4 works in concert with alpha and gamma subunits to mediate signal propagation. The complex modulates cyclic AMP levels and activates phospholipase C affecting various cellular responses. GNB4 interacts closely with proteins such as GNAS and ADCY1 forming part of the intricate signaling networks that drive cellular communication and response mechanisms.
Alterations or dysfunction of GNB4 can relate to certain neuropathological conditions. Mutations in the GNB4 gene have been linked to Charcot-Marie-Tooth disease which leads to peripheral neuropathy. This condition correlates with disruptions in GNB4's role in neuron signaling pathways. Additionally GNB4 interacts with proteins like PMP22 which when mutated also associates with this neuropathic disorder highlighting its involvement in these significant health conditions.
