RIPK5 also known as Receptor-Interacting Protein Kinase 5 is an important signaling molecule with a molecular mass of approximately 78 kDa. It belongs to the RIP kinase family characterized by a serine/threonine kinase domain. RIPK5 is expressed in a diverse range of tissues with higher expression levels noted in the brain and spleen. This distribution suggests a role in both neural function and immune response.
Biological function summary
RIPK5 functions as part of a larger protein complex involved in cellular stress responses. It contributes to the modulation of apoptosis and necroptosis two forms of programmed cell death. While RIPK5 itself does not initiate these processes its kinase activity modifies other proteins in the complex altering their function and the outcome of cell death pathways. This highlights RIPK5's integrative role in maintaining cellular homeostasis.
Pathways
RIPK5 participates in key signaling networks like the TNF receptor and NF-κB pathway. It interacts closely with proteins such as TAK1 and TRAF2 mediating downstream signaling cascades that affect inflammation and immune response. Through these interactions RIPK5 can influence cell survival differentiation and inflammatory responses impacting various physiological processes.
Research has linked RIPK5 to neurological disorders such as Alzheimer's disease and Parkinson's disease. Its association with proteins like tau and α-synuclein in these conditions suggests a potential role in neurodegeneration possibly through pathways that misregulate cell death or inflammatory responses. Understanding RIPK5's involvement in these diseases could provide insight into disease progression and opportunities for therapeutic interventions.
