The NRG2 protein also known as neuregulin 2 belongs to the neuregulin family and is involved in cell communication. It possesses a mass of approximately 44 kDa. NRG2 is present in various tissues with notable expression in the central nervous system heart and skeletal muscles. Its structure includes an epidermal growth factor (EGF)-like domain which enables interaction with other proteins on cell surfaces.
Biological function summary
NRG2 is involved in neural development and synaptic plasticity. It associates with ErbB receptor tyrosine kinases to mediate cellular responses. NRG2 forms a complex with these receptors to trigger downstream signaling pathways that support nerve cell growth and repair. As a ligand it selectively binds to ErbB3 and ErbB4 initiating pathways important for brain homeostasis and function.
Pathways
The NRG2 protein plays a significant role in the PI3K/AKT signaling and MAPK/ERK pathways. These pathways are essential for regulating cell survival proliferation and differentiation. Through the ErbB receptors NRG2 influences these signaling pathways intimately connecting it with other proteins such as AKT1 in the PI3K/AKT pathway and MEK1/2 in the MAPK/ERK pathway which further amplify its regulatory roles in cellular processes.
NRG2 is implicated in schizophrenia and peripheral nerve diseases. The interaction of NRG2 with the ErbB4 receptor has associations with schizophrenia where alterations in this signaling pathway may affect cognitive functions. In peripheral nerve diseases NRG2's role in neuron repair and maintenance can be significant linking it to ErbB3 in supporting nerve regeneration processes which becomes vital when addressing neuropathies.
