Protein reprimo (RPRM) also known as Reprimo is a small protein with a molecular mass of approximately 12.5 kDa. It does its mechanical function by regulating cell cycle arrest specifically at the G2/M checkpoint. RPRM localizes to the cytoplasm and is expressed in a varied range of tissues including the stomach and colon indicating its importance in gastrointestinal biology. It interacts with the cytoskeletal elements to exert its influence on cellular activities.
Biological function summary
RPRM acts as a tumor suppressor by inhibiting cell proliferation mainly through its G2 arrest function. It is not formally known to be part of a larger protein complex but it works closely with p53 a well-known regulatory protein and tumor suppressor to mediate its effects. RPRM expression helps prevent abnormal cell growth under stress conditions contributing to the maintenance of genomic stability.
Pathways
RPRM plays critical roles in the p53 signaling pathway and the cell cycle control pathway. In the p53 pathway RPRM aids in enforcing a cell cycle halt when DNA damage is detected working alongside other proteins such as p21 and GADD45. In the context of cell cycle control RPRM functions as an intermediary that facilitates the arrest of cell division in response to DNA stress signals collaborating with these proteins to enhance DNA repair mechanisms.
RPRM interactions significantly impact cancer particularly gastric cancer and colorectal cancer. The protein's reduced expression is frequently seen in malignancies where p53 pathway disruptions are apparent. In these cancers connections exist between the downregulation of RPRM and overexpression of Bcl-2 an anti-apoptotic protein where RPRM loss results in impaired apoptosis and uncontrolled cell survival. This association marks RPRM as a potential biomarker for understanding tumor dynamics in these types of cancers.
