VKORC1L1 also known as Vitamin K epoxide reductase complex subunit 1-like 1 is an important protein in the vitamin K cycle. It has a molecular mass of approximately 33.8 kDa. The protein is widely expressed including significant expression in the liver. Mechanically VKORC1L1 facilitates the reduction of vitamin K epoxide back to its active form which is a vital step for the regeneration of reduced vitamin K.
Biological function summary
The activity of regenerating reduced vitamin K is indispensable for the carboxylation of certain glutamic acid residues in several proteins. VKORC1L1 itself is not part of a large protein complex but its function strongly impacts enzymes that require vitamin K for activation. This biochemistry directly influences the synthesis of specific blood coagulation factors and some bone-related proteins which depend on carboxylation.
Pathways
The vitamin K cycle involves an essential role for VKORC1L1 mainly within the blood coagulation pathway and bone metabolism pathway. It interacts with multiple coagulation factors like Factor II VII IX and X which require vitamin K-dependent carboxylation for activation. In the bone metabolism pathway VKORC1L1 affects carboxylation of osteocalcin which is important for bone health.
The dysfunction or inhibition of VKORC1L1 can often lead to problems with blood coagulation. This protein links to conditions involving coagulopathy or bleeding disorders through its impact on blood clotting factors. Additionally VKORC1L1 influences osteoporosis through its effect on osteocalcin and bone density a condition where the balance of bone turnover is disrupted.
